So I upgraded the Cannondale from V-Brakes to disc. Do they make a screw in bolt to cover the hole after the brake studs are removed?

FWIW, threaded CO2 cartridges have the same thread. You can use the bosses for on-bike storage. [emoji41]

That’s funny and I would have never thought to try. I bought the more expensive inflator so that I could buy the cheaper c02 cartridges. Mine works with the thread less cartridges. I didn't have any issues removing them. My question was about covering the holes after they are out.
