Nope. I drilled out my i23 last year and have about a year of use on them. So far, no problems.
I've been drilling out rims to Schrader for something like 20 years, I've never had a rim fail because I drilled it.

i want to do the exact same thing. anyone know what size drill bit to use?
21/64" in SAE is the standard size for a Schrader Valve that we used when I was a bike wrench.

I drilled out both my i25 Freq rims. I have a drill press so it was super simple. Just keep that hole straight. I uses 949 brand schrader valves.
There is very little amount of material removed.
