Depends on the stem length and head tube angle of your frame.
For a given stem length and head angle the +6-degree stem will have more rise and less reach than the 0-degree stem. With -6 it is lower and has more reach. The longer the stem the greater the differences.
It depends, but it's noticeable. I have one of those Zoom adjustable stems, about 85 mm long. I put it on a new frame I built up to play with different angles. I noticed a huge difference going from 0 to ten degrees, was very surprised. Not very noticeable right away, but ater an hour or so my back was hurting with no rise, felt fine with 10 degrees. I'm running a six-degree stem now and it works well for me.
