Fellas,

My LBS has had my Bontrager Duster 29s off my 2013 Trek Rumblefish for the past week, and they said they aren't tubeless ready. I was told when I bought the bike they were. The LBS said they have tried multiple rim strips and nothing is working. I purchased new Spec tubeless tires to make this work. What is going on?

Did they try Stan's as well?
On the Stan's site:
NoTubes Rim Strip Finder
Select the second "Bontrager" in the first drop down
In the next, select 29 for size
In the third drop down, they have one for the Duster 29
This is what you come up with:
Bontrager , Size 29, Model Duster 29er

NoTubes Rim Strip: Flow
Product No.: RS0033

I can't think of anything else if they aren't TLR.

But that is saying it's for a TLR rim. My rim just says bontrager duster 29.
Duster's are 32 hole rim versions of the Rhythm wheelset. Same exact excursion, uses same rim strip.

Your LBS guys are tards, go somewhere else or order the parts I listed and do it yourself.

It only costs a few bucks to try gorilla tape and some sealant, the honest answer is that the majority of 'non-tubeless' stuff really can be quite adequate if you're not pushing hardware to its limits anyway.
